Description
The Human Amphiregulin ELISA quantitates Hu Amphiregulin in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu Amphiregulin. Principle of the method The Human Amphiregulin solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. L’amphiréguline (AR) se lie au récepteur de l’EGF (REGF) avec une moindre affinité que l’EGF. La forme d’AR sécrétée mature est un régulateur de croissance polypeptidique glycosylé résiduel à 84 acides aminés, qui est généré par transformation protéolytique d’un précurseur transmembranaire de 252 acides aminés. Sept ligands polypeptidiques différents, qui dérivent de gènes distincts, sont capables de se lier au domaine extracellulaire du REGF. Ces ligands incluent l’EGF, le TGF alpha, l’AR, le HB-EGF, le cripto-1, l’épiréguline et la bêta-celluline. Tous ces facteurs de croissance contiennent un domaine caractéristique de type EGF défini par six résidus de cystéine équidistants qui produisent trois boucles suite à la formation de ponts disulfures. La protéine AR a été localisée par immunohistochimie sur l’épithélium du côlon, de l’estomac, du pancréas, du sein et du placenta. L’AR est supposément surexprimée dans les cancers humains du sein, du côlon, de l’estomac et du pancréas.
