The Human TNFRSF6B (DcR3) ELISA quantitates Hu DcR3 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu DcR3. Principle of the method The Human DcR3 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. L’apoptose est induite par certaines cytokines, y compris le ligand TNF et Fas dans la famille des TNF, par leurs récepteurs contenant du domaine de mort. Plusieurs nouveaux membres de la famille TNFR, notamment DR3, DR4, DR5 et DR6, ont été récemment découverts et fonctionnent comme récepteurs de mort cellulaire. Deux récepteurs leurres, DcR1 et DcR2, ont récemment été identifiés pour concurrencer DR4 et DR5 pour leur liaison TRAIL de ligand. Un nouveau récepteur leurre a été découvert plus récemment et désigné DcR3 et TR6, respectivement. Contrairement à DcR1 et DcR2, DcR3 est une molécule soluble plutôt qu’une molécule associée à une membrane. DcR3 se lie à FasL et LIGHT et inhibe l’apoptose induite par FasL et LIGHT. La transcription de DcR3 est exprimée dans un certain nombre de carcinomes des poumons et du côlon et dans certains tissus normaux.
